Sorafenib, an anti-neoplastic drug, is prescribed for the treatment of liver cancer and advanced renal cell carcinoma.

Pharmacodynamics

The drug is a kinase inhibitor that inhibits malignant cell proliferation and intracellular levels of VEGFR, BRAF and CRAF. Such kinases are involved in the pathogenesis and replication of hepatocellular carcinoma and renal cell carcinoma.

Pharmacokinetics

After oral administration, the Cmax can be achieved within 3 hours. The drug is metabolized via the hepatic CYP3A4 pathway and UTI1A9 glucuronidation pathway. The drug is excreted mostly via the feces as glucuronide conjugates.

Sorafenib can cause malena, rashes, skin reactions, hemiplegia, paraplegia, malignant hypertension, easy bruising, rapid weight gain, paced heart beats, chest pain, goiter and irritation of skin, eyes and face. Some of the commonly reported adverse events of Sorafenib are hair thinning, weight loss, skin peeling, diarrhoea, nausea or vomiting, fatigue and numbness in the extremities.
