Pazufloxacin, a form of fused tricyclic quinolone and broad-spectrum antibiotic, is prescribed for the treatment of several infections due to Gram-positive and Gram-negative organisms including anaerobic and drug-resistant strains. Pazufloxacin is indicated for the treatment of the respiratory tract infections including lung abscess and pneumonia, endometritis, prostatitis, cystitis, pyelonephritis, liver abscess, intra-abdominal infections, peritonitis, cholecystitis and other secondary infections due to injuries, post-surgical wounds and burns.

Pharmacodynamics

Pazufloxacin selectively inhibits DNA gyrase and topoisomerase IV that exerts broad-spectrum antibiotic activity. The drug molecule exhibits DNA antagonism with multimodal action to lower drug-resistance. The efflux mechanism of drug resistance was not reported in Pazufloxacin treatment.

Pharmacokinetics

After intravenous administration, the drug achieve Cmax with in 30 minutes and widely distributed in the pleural fluid, phlegm, lungs, biliary tract, CSF and genital organs. The terminal half-life of Pazufloxacin is 1.65-1.88 hours. Pazufloxacin is mostly excreted as unchanged drug via the urine with mild or negligible rate of systemic accumulation.

Dosage

For Infections (Adults):
The recommended adult dose of Pazufloxacin injection is 500 mg, twice daily. The dosage should be administered as an intravenous fluid over 30-60 minutes. The dosage can be adjusted based on the patient’s age, weight and clinical response. The maximum duration of Pazufloxacin therapy should not exceed 14 days.
